Abstract

There is an increasing interest in the utility of dental pulp stem cells (DPSCs) for dental tissue regeneration. The purposes of this study were to isolate, to characterize and to lead cellular differentiation of stem cells from human dental pulp from extracted premolars of young patients. The stem cells isolated in cellular cultures generated clonogenic cultures after 5 weeks, presenting elongated and flat cells. This cells expressed mesenchymal stem cell markers (STRO-1 and CD-44) as shown by immunocytochemistry essay and they could differentiate in vitro into osteoblastic-like lineage. This differentiation was evaluated by RTPCR and by immunocytochemistry and the results revealed that bone sialoprotein and osteopontin, two very important extracellular matrix protein, were expressed in the induction cultures and it also confirms the osteoblastic-like phenotype. In conclusion, cells from dental pulp with stem cell characteristics were isolated and the present study suggests that stem cells from human dental pulp may potentially be used as a cellular therapy in the tissue engineering area.
